Objective

To synthesize 42 Ugi products in parallel using Mettler Toledo's MiniMapper and MiniBlock system. Six experiments will be repeated in order to verify the consistency.

Discussion

It is hard to tell from the images if there is precipitate - does physically inspecting the tubes indicate that there is more precipitate than appears in images? - I did not see anymore ppts than seen from the pics.

Is there any reason to expect the Ugi products resulting from the reactions with significant precipitates to be insoluble in CDCl3? What other chemistry might be going on with the acid you're using to give insoluble products?

Conclusion

The results are not meaningful because the liquid handler was possibly not properly programmed.

12:30 The reaction tubes have been on the shaker until now...stopped the shaker and obtained a picture..3rd day.

10:30 Vacuumed off excess solvent from the reaction tubes.

11:10 Removed from the shaker and vacuumed off solvent form the reaction tubes, and added methanol (500uL) for a second wash and set the miniblock on the shaker again.
11:20 Removed from the shaker and vacuumed off solvent form the reaction tubes- left the reaction tubes connected to the house-vh:ac for drying.
18:00 Removed from vacuum and weighed the tubes.... just D:7 and E:10 contained some significant amount of precipitates.
19:00 The precipitates from D:7 and E:10 remained insoluble in CDCl3.
20:00 These ppts were left under vacuum to evaporate CDCl3.

10:20 Tried to dissolve the ppts obtained in DMSO-d6.D:7 and E:10 remained sparingly soluble in DMSO-d6 too....however got a HNMR of E10 - (pls show NMR)......after getting an HNMR added D2O (500uL)..still insoluble...
